Single-molecule imaging has been recently developed as a novel technology, which allows us to monitor behaviors of single biological molecules such as proteins.In the single-molecule imaging measurement, practical resolution hasbeen defined by the Rayleigh limit in a deterministic way. In a real situation, however, there are a number of probabilistic events, e.g., a probabilistic fluctuation of number of photons, or a probabilistic scatter of photons by the diffraction.
In this article, we consider such a probabilictic fluctuation and distinguish the number of fluorescent molecules being one from that being two based on the likelihood ratio test, which is known to be the most powerful statistical test. This framework also enables us to consider the condition for achieving the best observation resolution based on calculating the error for resolution power. |
