The ionic dynamics depending on the nervous activity recently attracts attentions of many researchers due to the rapid progress of Ion-Imaging techniques.
However, there are several problems to be addressed in the ion-imaging techniques.
In this paper, we developed statistical estimation methods for calcium ion dynamics from noisy fluoresce signals. First, we composed a state-space model describing calcium ion inflow kinetics, binding kinetics of calcium buffers and calcium indicators, and optical measuring devices for detecting fluorescent signals.
Then, we derived an algorithm based on the belief propagation method for estimating calcium ion concentrations, and the EM algorithm for estimating model parameters.
We numerically verified the efficacy of our algorithms by applying to artificial data. Our algorithm proposed here makes it possible to estimate not only intracellular calcium ion concentration but also calcium ion inflow through the cell membrane and Endoplasmic Reticula (ER) membrane et al.
It is expected that the understanding for the molecular mechanisms such as the calcium channels and Ca store can be promoted by our method, since variation of calcium inflows give the significant information these molecular mechanisms. |
