A sleep-wake rhythm is a biological rhythm with a roughly 24-hour period and widely observed in animals. This rhythm consists of two cycles. One is a long cycle with state transitions between wake state and sleep state and the other is a short cycle with state transitions between rapid eye movement (REM) sleep state and non-REM sleep state within a sleep state. It has been reported that a neuropeptide, Orexin, plays an important role in regulating the sleep-wake rhythm, and the deficient of Orexin causes an irregular sleep-wake rhythm similar to a sleep disorder called narcolepsy. It is, however, not fully understood how Orexin controls the sleep-wake rhythm. Recently, while several mathematical models describing the role of Orexin in regulating the sleep-wake rhythms have been proposed, any previous model with the dynamics of an Orexinergic neuronal population couldn't reproduce three-state transition among wake state, REM sleep state, and NREM sleep state. Fulcher et al. (2014) introduced a mathematical model which describes dynamics of Orexinergic neuronal population and shows two state transition between wake state and sleep state. On the other hand, Diniz Behn et al. (2013) introduced a model called MI model which describes another state transition between REM sleep state and NREM sleep state. In this study, we proposed a mathematical model by combining Fulcher et al.'s Model and MI model of Diniz Behn et al.. We demonstrated that our model can reproduce the features of Orexin deficiency obtained by previous physiological experiments: increasing the number of occurrences of REM sleep, inducing the frequent transition from wake state to REM sleep state, decreasing appearance time and duration of wake state, increasing appearance time and duration of REM sleep state. |
