Multi-electrode arrays (MEAs) are widely used as cellular interfaces for large-scale neuronal recording. MEAs are broadly classified into flexible sheet-type surface electrodes and rigid needle-type penetration electrodes. While the penetration electrode can directly measure spiking activity of neurons even in the deep brain region, it is known to be invasive for the brain tissue. Surface electrodes are relatively low invasive but only measures local field potentials (LFPs) on the surface. If the relationship between the signals measured by the surface electrode and those measured by the penetration electrode can be clarified, it may be possible to estimate neural activity in the deep brain region only from minimally invasive surface electrode measurements. In this study, we investigated the relationship between neural responses at the surface and deep brain regions by simultaneous measurement by surface electrodes and penetration electrodes in the auditory cortex of rats. Time-frequency analysis, which can quantify the neural responses in specific frequency components, revealed a strong correlation between the neural response at the surface electrode and that at the penetration electrode, specifically in the Hγ band (70-170 Hz). Furthermore, the Hγ band responses were found to reflect the frequency and sound pressure level of the sound stimulus. These results indicate the possibility to estimate auditory neural responses in the deep brain only from low-invasive surface electrode measurements.
