(in English) NMDA receptor (NMDAR) and Ca2+/calmodulin dependent protein kinase II (CaMKII) are important to induce long-term potentiation (LTP). We clarified so far the application of NMDAR antagonist, AP5 or CaMKII inhibitors, KN-93 decreased the frequency of bicuculline (BIC)-induced epileptic discharges (BIED) when those drugs were pre-applied before the induction of the discharges. To determine whether the decrease of the frequency is induced by the suppression of the NMDAR- and CaMKII-dependent LTP, we measured the excitatory synaptic plasticity in the appearance of BIED. The epileptic discharges were recorded from the guinea-pig hippocampal slices (500 µm thick) using a glass micropipette when BIC, a GABAa receptor antagonist, was applied to the slices. Evoke population EPSP (pEPSP) at CA3-CA3 synapses was measured when schaffer collateral/commissure fibers were antidromicaly stimulated. Evoked pEPSP was potentiated between 210 to 240 min after BIC was rinsed and BIED disappeared. The potentiation was not measured by the application of AP5 or KN-93 with BIC. Moreover, to examine whether the strength of the excitatory synaptic transmission affects the frequency of BIED, AMPA receptor antagonist, CNQX was applied to BIED. It decreased the frequency on the concentration-dependent manner. Theses results suggest that the BIED induce the NMDAR- and CaMKII-dependent potentiation and the degree of the potentiation can determine the frequency of the discharges.
