Ca2+ channels play pivotal roles for neuronal functions. The precise distribution of the high-voltage activated (HVA) calcium channel subtypes (L-, N-, P-, Q-, R- types) and low-voltage activated (LVA or T-type) calcium channel in neuronal soma membrane has been confirmed by using immunohistochemistry as well as electrophysiological approaches. Some calcium channel subtypes such as P-type were found to be linked mainly to dendrite localisation. However, the functional specificities of the subtypes which distribute on the small nerve endings (Φ: less than 1μm) are not known so far. In the present study, the single neurons mechanically dissociated from the sacral commissural nucleus (SDCN) of rat (7-15 days old) spinal cord were used. These single neurons, namely ‘synaptic bouton’ preparations, are attached with many functional nerve terminals. One of single boutons was selectively stimulated (5-10μA,0.2 Hz,100μsec of duration) by using ‘focal stimulation’ technique. The evoked inhibitory glycinergic postsynaptic currents (eIPSCs) were isolated pharmacologically by blocking the glutamatergic and GABAergic inputs with CNQX/D-AP5 and bicuculline, respectively. Successively, specific HVA and LVA Ca2+ channel subtype blockers (ω-con GVIA, ω-Aga IVA, nifedipine, efonidipine) and non- selective blocker were examined on glycinergic eIPSCs to distinguish each HVA and LVA Ca2+ channel subtypes. In addition, obtained results were compared with those of HVA Ca2+ channel subtypes distribution of GABAergic nerve endings on hippocampus CA1 neuron. |
