Estimation algorithm using direct search method in PET neuroreceptor quantification

(in English) In a compartment model analysis using positron emission tomography (PET), some problems arises in non-linear estimation: misconvergence, dependence in estimation results on a initial values and calculation time. This study aimed at investigating a direct search method. In the algorithm, feature space on a time history of radioactivity concentration in tissues was composed using a measured time history in arterial plasma and a set of rate constraints in the model that covered physiologically expected range. The space was fluctuated by the noise observed in ROI-based PET study. The dynamic PET data were projected onto the feature space, and then clustered using the Mahalanobis distance. The method was validated with simulation based on ROI studies for [11C]TMSX. Especially, the kinetic parameter K1 and a total distribution volume, VT , were estimated well. If VT was fixed at this estimate and the algorithem was applied again, a performance in model estimation on a neuroreceptor density was approximately 20% of quartile deviation that was 40% without the constraint.
