For intractable neural diseases, clinical trials and basic researches on the compensations/prostheses of the neuronal functions by using exogenously delivered electric stimulation have been widely performed in recent years.In the case of invasive neural prostheses,it is critical to use the necessary, but minimum amount of the electrical current/charge to stimulate the target neurons, in order to guarantee their safety.However,in previous clinical studies, the rectangular pulse of stimulation had a few tens of $\mu$A in amplitude and a few hundreds of μsec in duration; these resulted in roughly 100-1000 times larger than the electrical charge required for the firing of a neuronal impulse under normal physiological condition. Thus, those stimulation seem not to be optimal about its electrical charge, waveform and the position of stimulation electrode.For the next generation of the neuronal prosthesis achieving a finer resolution at the single-cell level, it is required electrophysiological analyses in more quantitative manner. In this study,we performed computer simulations using biophysics neuron models with three-dimensional morphologies in order to quantitatively analyze the mechanism of the neuronal excitatory responses to extracellular current stimuli and the effects of stimulation parameters on the evoked responses. |
