ラット海馬CA1錐体細胞において、直流細胞外電場に対する膜電位応答を実験的に観察したところ、基底樹状突起-細胞体部位では遅い時間経過をもつ単相性の応答が、尖端樹状突起部位においては速い成分と遅い成分をもつ二相性の応答が生じる。また同様に、交流細胞外電場に対する膜電位応答では尖端樹状突起部位において周波数依存的な応答を呈する事が膜電位イメージング法によって観察されている。これまで、これらの特徴は一端の膜抵抗値が低いリーキー単一ケーブルモデルによって再現可能である事が分かっている。また、曲がりや分枝といった神経細胞の形態的特徴によっても実験的特徴が再現可能であるという仮説の下、形態的特徴をもったケーブルモデルを用いて、直流細胞外電場に対する膜電位応答を数値的にシミュレートし、適切な形態によっては部分的に実験的特徴を再現可能である事が分かっている。そこで次に、形態的特徴をもったケーブルモデルを用いて、交流細胞外電場に対する膜電位応答を検証したところ、周波数依存的な特性を呈する事が分かった。 |

Previously we reported that, in response to an applied extracellular DC field, the membrane potential near the soma changes with slow time course, and the potential in the apical dendrites shows biphasic change with with fast and slow time course. For AC field, the potential in the apical dendrites shows frequency dependent changes. In our recent modeling studies, we have shown that a leaky cable model with low membrane resistivity on one end mimics these behaviors. As an alternative account for the experimental results, we tested the impact of the morphology of the cells for DC field induced responses, and found that the morphology can also be responsible for the behavior of the cells. Here we test the impact of the morphology for AC field by performing compartmental model simulations, to find that the morphology also can account for the frequency dependent behavior of the dendrites. |
