Glutamate is released from injured neurons and astrocytes caused by severe ischemic stress. A marked elevation in the concentration of extracellular glutamate (Glu) induces excessive neuronal depolarization, resulting in acute neuronal death in the ischemic core region. In addition, Glu diffused from the ischemic core possibly induces delayed neuronal death in the surroundings of the core region (ischemic penumbra). Here we developed a new in vitro model of brain ischemia by focal photolysis of caged glutamate. A caged glutamate is one of the “caged compounds” activated by irradiation with a pulse of ultraviolet light. This method makes it possible to increase the concentration of extracellular Glu in the focal flashed area. Focal photolysis of a caged Glu resulted in the induction of acute neuronal death in the photolytic-flashed area (FA) and delayed neuronal death in the region surrounding the FA (SFA) in neuron/glia co-cultures. |
