Signal transduction mechanisms for the regulation of ATP release during ischemia in cardiomyocytes

(in English) A variety of cellular mechanisms are involved in the release of intracellular ATP in cardiomyocytes. It is reported that the maxi-anion channel is the major pathway for ATP release in the early stage of ischemia, while ATP is also released through hemichannels during ischemia. Recent findings have revealed that ATP works as one of the extracellular signaling molecules responsible for intercellular communications. However, functional relationships not only between ATP release via maxi-anion channels and that via hemichannels, but also between the quantity of ATP released and ischemia/reperfusion injury in cardiomyocytes are unknown. In this study, we aimed at clarifying the intracellular signal transduction system regulating ATP release during ischemia in cultured cardiomyocytes. The present study suggested that there existed a negative-feedback regulatory mechanism for the maxi-anion channel-derived ATP-induced suppression of further ATP release via hemichannels during ischemia.
