The observation data of the calcium imaging techniques are indirect information about the intracellular calcium ion concentration. To quantitatively estimate the calcium ion concentration, we have to calibrate calcium fluorescence data. We applied the state space method to solve this calibration problem. We constructed the model of calcium dynamics containing discontinuous changes with the line process and the second-order trend model. We implemented our state-space model with the particle filter and performed Bayesian estimation for calcium ion concentration. We verified its effectiveness by applying it to articial and real fluorescence data. |
