We have been developing a liquid-phase immunoassay technique for the detection of biological targets using SQUID and magnetic markers. The performance of the magnetic detection method depends on the property of the magnetic markers, as well as the sensitivity of the measurement system. At present, however, development of the magnetic markers that are suitable for magnetic immunoassay is not established. Since magnetic markers are usually composed of aggregated magnetic nanoparticles, their properties are significantly affected by the degree of aggregation. Therefore, we obtained the distribution of the magnetic moment m and the Neel relaxation time N of the aggregated particles included in the markers through the analysis of magnetization (M-H) and AC susceptibility measurements. We also determined the relationship between m and N. Using the obtained result, we selected the magnetic marker that is suitable for magnetic immunoassay. We also improved the measurement system using the HTS SQUID, and demonstrated the high sensitive detection of biological targets with the liquid-phase immunoassay. |
