Electrochemical imaging techniques including scanning electrochemical microscopy (SECM) and local redox cycling-based electrochemical (LRC-EC) chip device have been applied for quantitative evaluation of alkaline phosphatase (ALP) activity of mouse embryonic stem (ES) cells to judge the state of undifferentiation of the cellular samples. Our techniques allowed rapid and high-throughput measuring procedures for ALP activity of stem cells which remarkably improved the invasiveness against the undifferentiated cellular samples. The ES cells and embryoid bodies (EBs) that have been used for ALP assay were found to be re-usable for further cultivation to induce differentiation into cardiomyocyte-like cells. We also mention about background researches done by us and other groups including non-invasive measurement of respiration activity of embryos and EBs for quality control. |
