Investigation into directional electron transfer of cytochrome c3 by electrochemical measurement

(in English) Cytochrome c3 is a redox protein containing four hemes in one molecule and works as an electron mediator in vivo. There is intense interest in experiments which reveal, at molecular level, how electrons go into and out of cytochrome c3. Here we report that specific heme which roles in an entrance or an exit during electron transfer, that is direction of electron transfer through cytochrome c3, is clarified by electrochemical quartz crystal microbalance (EQCM) measurement.
In vivo, cytochrome c3 accepts/donates electrons from/to hydrogenase, which is catalyze proton reduction and hydrogen oxidation. In this study, electron transfer between cytochrome c3 and hydrogenase was measured electrochemically, and the electron transfer complex formation was investigated by frequency change of the quartz crystal at the same time as electrochemistry.
