It has been reported with multicellular calcium imaging methods that calcium transients are entrained to the extracellular AC field in CA1 region of rat hippocampal slices. In this study, we apply statistical phase analysis method for the calcium imaging data and analyze the entrainment of calcium transients to the extracellular AC field. We developed a statistical method consisting of the following process steps: automatic detection of the fluorescence intensity changes of individual cells, phase estimation from the fluorescence intensity changes and parameter estimation of phase equation. By numerical experiments on artificial data, we show that our method can approximately estimate natural frequencies and coupling coefficients of the phase equation well in the case of weakly coupling. We then apply the method to the imaging data of rat hippocampal slices to how that our method can estimate natural frequencies and coupling coefficients to the extracellular AC field. |
