3D Superresolution of Microscopic Images based on Variational Bayesian Inference via Chebyshev polynomials approximation

(in English) Optical microscopes are used to elucidate changes in cellular functions mediated by morphological changes of cells in vivo. Although only 2D images can be obtained by single scans, depth information of the measurement target can be acquired by scanning and integrating multiple images with different focal depth. However, we cannot obtain high spatial resolution 3D images in some cases, because of the necessity of observing wide region, fluorescence loss due to photobleaching, and so on. In this study, we propose a statistical image processing method to estimate super-resolved depth and intensity maps based on Bayesian inference; as a prior knowledge, we use a model of the point spread function (PSF) and label-based information of the target. To achieve an efficient computation of the Bayesian inference, we use Chebyshev approximation of the PSF and numerical integration. In simulations using artificial images of a neuronal cell, our method can estimate both of the super-resolved image and the object's depth from multiple 2D observed images.
