Measurement of change in endogenous serotonin levels by PET dynamic analysis

(in English) The neurotransmitter, such as dopamine and serotonin, is related to depression, Parkinson disease, and other psychiatric or neurologic disorders. In the measurement of neuroreceptors using positron emission tomography (PET), the binding of administered radioligand to the receptors often competes with endogenous neurotransmitters. Therefore, an increase or decrease of the neurotransmitters in the synaptic cleft can be estimated from change in the binding of radioligand to the receptors. In this study, we targeted to the quantification of the change in the serotonin level, time-activity curves obtained by the PET dynamic scan were simulated using a compartment model including the administered radioligand and endogenous serotonin concentration. From these curves, we evaluated the analysis method for estimating the change in endogenous serotonin level, and validated the proposed method by small animal PET scans.
