Recently, fetal brains could have been imaged by magnetic resonance imaging (MRI) and analyzed in detail morphologically. Those images were quite fine, however, obtained by isolated brain samples, and using MRI system for experimental use therefore it is difficult to obtain enough data for fetal brain research. To facilitate the human fetal brain imaging, we tried to establish the protocol suitable for whole-body human fetuses using MRI system with 3T magnet, for clinical use. For magnetization-prepared rapid gradient-echo (MP-RAGE) sequence, no treatment were not required for fetuses in formalin solution to obtain fine images; but less signals could be obtained from the specimens in T2-weighted and diffusion-weighted imaging (DWI). To improve signals from water molecule, the fetuses were soaked in phosphate buffered saline for 3-14 days and the T2 and DWI signals were improved after the substitution. The specimens with long-term preservation in formalin solution were available for T2 and DWI as well as MP-RAGE, but tractography was not obtained from it. As next attempt, small quantity of formalin was injected into brain of new sample and the sample was soaked in PBS before imaging. As a result, clear color map and tractography was obtained by using the sample. It is required to consider method to obtain strong signal of diffusion tensor after long-time preservation. |
