Texture profiling for local property analysis of induced pluripotent stem cells colony

(in English) The induced pluripotent stem (iPS) cells represent an aggregate characteristic appearance in a form of colony. Colonies having an ill-conditioned appearance are selectively removed in the process of culturing to produce high-quality iPS cells, whereas culturing iPS cells is technically difficult in itself. Presently, such a quality control in iPS cell culture depends on experiment and intuition of an operator, which hampers the industrialization because of the inefficiency. In this study, we propose a method to objectively and quantitatively classify the state of iPS cell colony using a support vector machine based on its texture features. We evaluate the efficacy by comparing the segmentation result by the proposed method and the cell evaluation utilizing immunological dyeing method.
