Recently, research fields called bioimage informatics for obtaining quantitative data of cell images by a computer has grown up. In order to analyze cell images, it is first necessary to segment the cells from the background. Conventionally, a threshold-based binarization method is used for segmenting a cell image, but it is sometimes difficult to successfully extract it based on the statistical properties of the image. Therefore, we introduced an index of a stable boundary focusing on the geometrical structure of the image and proposed a binarization method that selects a scale whose boundaries are all surrounded by stable boundaries. However, with the conventional method, since a scale is selected in which all boundaries of the object are covered with stable boundaries, there is a problem that it can be cut out only with a coarse structure. In this article, we focus on the hierarchical structure of the object in the scale space and propose a highly accurate binarization method independent of spatial stability of different boundaries for each scale. |
