It is a well-known fact that underwater microorganisms can be inactivated through the administration of ultrasonic energy. We have reported that the main reason behind the deactivation is cell disruption via the ultrasound treatment of samples such as algae and fungi. In addition, we observed that the mechanical resonance of the cell is responsible for its disruption. In this study, two species classified as Diatoms (Chaetoceros gracilis, Chaetoceros sp.) and Prasinophyceae (Tetraselmis sp.) were used. The average particle sizes were about 4.0 μm, 4.5 μm, and 8.7 μm, respectively. Three species of algae were irradiated by ultrasound with different frequencies of 20 kHz, 400 kHz, 1.0 MHz, 2.4 MHz, 3.4 MHz, and 4.4 MHz to investigate the disruption mechanism of algae. As a result, algae with larger particle sizes tended to be disrupted at lower frequencies. In addition, under the same acoustic power, for each frequency, it was difficult to observe cell disruption in Tetraselmis sp. and it did not change as much as the two species of Diatoms. It is observed that the area deformation of microorganism’s cell membrane by ultrasonic cavitation is important for cell disruption. |
