Expression of c-Fos in brain regions associated with the promotion of sleep-wakefulness and feeding induced by intracerebroventricular administration of ghrelin

(in English) Ghrelin is a promoter of sleep-wakefulness and food intake in rodents. Since few studies examined its brain effects, we investigated the effects of intracerebroventricular (i.c.v) administration of ghrelin on sleep-wakefulness, food intake and c-Fos expression of some brain regions in rats. Protein c-Fos was used as a molecular maker of neuronal activity in brain sites activated by ghrelin. The results show that i.c.v ghrelin promotes wakefulness and increases c-Fos expressions in serotonergic (5-HT) neurons of the dorsal raphe (DR) and in noradrenergic (NA) neurons of the locus coeruleus (LC). The results also indicates that i.c.v ghrelin may increase food intake and the number of c-Fos-positive orexinergic (OX) lateral hypothalamic (LH) neurons. We suggest that the promotion of wakefulness by ghrelin is mediated possibly through the activation of the DR 5-HTergic and LC NAergic neurons, while the orexigenic effect of ghrelin may be due to the activation of the LH OXergic neurons.
