A Neural-Glial Model of Lateral Habenula Circuits for Cognitive Distortions in Depression

(in English) Depression is associated with biased cognition, such as heightened responses to negative events and reduced sensitivity to positive events, which may contribute to symptom persistence. Experimental studies suggest that chronic stress elevates lateral habenula (LHb: lateral habenula) activity and that neuron--astrocyte interactions in LHb are involved in depressive-like behaviors. However, a compact dynamical framework that links these findings to conditions under which circuit activity fails to recover remains limited. We constructed a minimal LHb--locus coeruleus (LC: locus coeruleus) rate model augmented with an astrocytic state variable $Y$ and a coupling modulation $g(Y)$. A brief stress-like rectangular input can shift the system into a sustained high-activity state (High Plateau), and the maximal modulation $g_{max}$ controls a qualitative transition from transient to persistent responses. A two-dimensional parameter sweep further identified the region in which High Plateau occurs. These results provide a basis for interpreting stress--LHb--astrocyte positive feedback as a state switch that becomes difficult to reverse.
