We successfully detected silicon-binding proteins such as Si-tagged protein A, green fluorescent protein (GFP) and luciferase attached to OH-terminated SiO2 and H-terminated Si surfaces by surface potential measurements using an AFM/Kelvin probe technique. We also observed significant change in electrical conductivity of lightly phosphorous doped poly-Si films after attachment of Si-tagged proteins. The conductivity of the poly-Si films is increased from 1.0x10-5 S/cm to 3.7x10-4 S/cm with the attachment of protein A. It is suggested that field effect passivation of unoccupied gap states in polycrystalline Si film increased the film conductivity. These results will open up the fabrication of a sensing device for locally ionized biomolecules. |
