For measuring concentration of free calcium ([Ca2+]i) inside living cells, indicator dyes including fura-2 are widely used. It should be appreciated, however, that such indicator dye is a divalent metal ion chelator which changes its fluorescence properties and intrinsic intracellular Ca2+ dynamics. Because, as chelators, indicator dyes may influence the quantity of interest, the Ca signal, it is important to know its influence on intracellular Ca2+ dynamics quantitatively. In this study, to estimate [Ca2+]i, for the first step, we have proposed a statistical inference method in which [Ca2+]i is inferred by measurements of fluorescence signals and Ca2+ current that inflows thorough cellular membrane. Firstly, we constructed a state-space model describing the dynamics whose state variables are concentrations of calcium ion, calcium buffer, and calcium indicators. Then, its self-organizing state-space model was defined by augmenting a state vector with unknown parameters of the original state-space model. In the model, unknown parameters were estimated equally as the original state vector. Next, we applied recursive Bayesian estimation to infer a set of the state vector and the unknown parameters from a set of the observation vector. To calculate the recursive Bayesian estimation, we used a sequential Monte-Carlo method, which is referred to as a particle filter method. To verify effectiveness of the proposed method, we carried out numerical experiments for a test model with known parameters. As the result, we show that the proposed method could estimate the time course of [Ca2+]i and the indicator dye concentration. |
