Basic study of animal test alternative using cultured nerve cell for the safety assessment of neural interface

(in English) Direct neural interface is intensively developed to artificially reproduce a lost sensation. The safety of it is evaluated by animal experiment. However, the evaluation using experimental animal has not only problems of high economic and temporal cost but also ethical issues. Therefore, we are developing an animal test alternative that can evaluate the influence of stimulation to nerve cells in vitro. In this study, we investigated the period during which neural activity of nerve cells (NG108–15) cultured on multi electrode array is maintained. Using culture method with medium circulation, the spontaneous-spike activity was able to be obtained until 62.5 hours. Furthermore, the record of nerve activity over 9 days was achieved by culture method with medium exchanging.
